Protein posttranslational modification(PTM) is an important term that refers to biochemical modification of specific amino acid residues in target proteins, which immensely increases the functional diversity of the proteome. The range of PTMs includes phosphorylation, acetylation, methylation, ubiquitination, SUMOylation, glycosylation, and several novel PTMs.
PTM has been considered as a potential pharmacological intervention in preclinical research and clinical trials, promising new therapeutic strategies for disease treatment. Families of drugs targeting protein modification enzymes include kinase agonists and inhibitors, HDAC inhibitors, histone methyltransferase inhibitors, and others. Table 1 shows the current clinicaltrials of metabolic diseases by targeting protein modification.
